Abstract

PURPOSE: Aerobic exercise plays an important role of preventing type 2 diabetes mellitus induced by high-sugar and high-lipid fodder. Some reports showed that there was a association of body iron stores and T2DM risk. The iron deposition and iron-induced oxidative stress contribute to the pathogenesis of T2DM. It is not known whether the decreased oxidative stress and insulin resistance by exercise related to iron metabolism. The research aimed to illuminate the regulatory mechanism of exercise on iron metabolism during preventing of T2DM. METHODS: 24 male SD rats,12-month old, were randomly divided into three groups, the control group (CG, fed by standard diet and kept sedentary), the high-sugar-lipid fodder Group (HG, kept sedentary) and Exercise with high-sugar-lipid fodder Group (HE, run on treadmill at 15m/min, 45min/d, 6d/w, 5W). The content of serum ferritin and insulin were examined by ELISA kits. Serum iron, hepatic iron, plasma glucose and the activity of SOD in liver were examined by reagent kits. The iron transporter hepatic ferroportin 1(FPN1)were examined by Western Blot. RESULTS: (1) The plasma glucose of HG were obviously higher than those in CG and HE(CG: 5.83±0.27 mmol/L, HG: 13.42±0.80 mmol/L, HE: 8.88±0.76 mmol/L, P<0.01). The serum insulin of HG were increased compared with CG and HE(CG: 31.48±2.01μU/ml, HG: 45.62±2.87μU/ml, HE: 35.18±2.94μU/ml, P<0.01). (2) The content of serum ferritin (SF) in HE was higher than that of CG, while which was lower than that of HG (CG: 240.50±12.10 pg/ml, HG: 337.43±28.14 pg/ml, HE: 294.28±24.59 pg/ml, P<0.01). The total hepatic iron of HG was significantly increased than that in CG and HE (CG: 37.64±2.64μmol/g, HG: 45.67±7.99μmol/g, HE: 35.08±5.46μmol/g, P<0.05). (3)The activity of hepatic SOD of HE was significantly increased compared with HG (HG: 36.49±6.59 U/mg, HE: 45.35±3.54 U/mg, P<0.01). (4)The expression of hepatic FPN1 of HE was significantly increased than that of CG and HG( P<0.01). CONCLUSIONS: (1) The aggravated iron overload and oxidative stress were increased after the High-sugar-lipid fodder, which induced T2DM.(2) The iron released was promoted by aerobic exercise, which remitted iron overload in tissue and elevated the activity of enzymatic antioxidants. As a result, the oxidative stress and plasma glucose were decreased after exercise.
