Abstract

The effects of 1 nM 17β-estradiol, 1 nM 2,3,7,8-tetrachlorodibenzo- p-dioxin (TCDD) and their combination on the formation of intracellular cytosolic 52- and 34-kDa proteins was determined using a commercially available cathepsin D radioimmunoassay procedure (ELSA cath-D™ kit) which recognizes both the 34- and 52-kDa proteins. In control (ethanol-treated) cells, the 52-kDa protein levels were 305 ± 49 fmol/mg protein (100%) whereas in cells treated with 1 nM 17β-estradiol, 1 nM TCDD or their combination the levels were 1223 ± 142 (401%), 478 ± 54 (150%) and 227 ± 33 (74%). These results confirm that TCDD inhibits the 17β-estradiol-induced intracellular formation of the 52-/34-kDa proteins and further extends the antiestrogenic properties of this compound.
