Abstract

Objective To investigate the effects and mechanisms of Quercetin on PDGF-BB induced proliferation and migration of human airway smooth muscle cells(HASMCs).Methods HASMCs derived from hurran bronchus were divided into six groups:control group,PDGF-BB group,PDGF-BB and Quercetin intervention group,Quercetin group,U0126 group,PDGF BB and U0126 intervention group.Proliferation was measured by MTT.Migration was examined by Transwell test.The protein expressions of p-ERK and eyclin D1 were determined by western blot analysis.Results PDGF-BB (20μg/L)significantly induced proliferation and migration of HASMCs as compared to control.Quercetin(20~80μmol/L) inhibited the HASMCs proliferation and migration in response to PDGF BB (20μg/L).The phosphorylated protein levels of ERK and cyclin D1 in PDGF-BB group were higher than those of the control group (P<0.05),but decreased in PDGF-BB and Quercetin intervention group compared with control group (P<0.05).Conclusions It appears that quercetin can directly inhibit PDGF-BB induced HASMCs proliferation and migration via the modulation of the ERK/cyclin D1 pathway.
