Abstract

Objective To investigate the effect of small interference RNA(siRNA)silencing c-myc on the biological behaviors of human pancreatic cancer cell line SW1990. Methods The c-myc gene expression was knocked down in human pancreatic cancer cell line SW1990 by using c-myc siRNA.The expression level of c-myc was detected in pancreatic cancer cell line SW1990 at mRNA and protein levels by using real-time reverse transcription-polymerase chain reaction(RT-qPCR)or Western blotting.The cell proliferation was measured by methyl thiazol tetrazolium(MTT)method.The flow cytometry was used to detect the apoptosis rate of SW1990 cells.The migration was observed by using Transwell cell migration system. Results The cmyc siRNA could down-regulate the expression of c-myc mRNA and protein in SW1990 cells remarkably. the mRNA level of c-myc siRNA groups, which was(0. 263±0. 048), decreased compared with the siRNA-NC groups(0. 970±0. 012)and control groups 1 significantly.The proliferation of SW1990 cells was decreased in c-myc siRNA groups significantly. The apoptosis rate in c-myc siRNA group[(19. 90± 2. 09)%], was significantly elevated as compared with the siRNA-NC group[(4. 93±0. 25)%]and control group[(4. 40±0. 34)%].The c-myc siRNA could inhibit migration of SW1990 cells(34. 3±1. 2 vs.68. 3± 5. 8,72. 3±1. 2)as compared with the siRNA-NC group and control group. Conclusion The c-myc siRNA can down-regulate the expression of c-myc in SW1990 cells and decrease the proliferation and migration, promoting the apoptosis of SW1990 cells.
