Abstract
Human proximal tubule epithelial cell line,HK-2 cells,were cultured with various concentrations of estradiol or testosterone for 48 h.Human urate transporter(hUAT)Mrna was detected by realtime quantitative PCR.HUAT mRNA levels were up-regulated by estradiol(10~(-12),10~(-10),10~(-8),10~(-6) mol/L)in a dose-dependent manner(relative expression median were 1.03,1.05,1.67,and 1.87),while down-regulated by testosterone.The relative expression median in 10~(-7),10~(-6),10~(-5),10~(-4) mol/L testosterone were 0.56,0.34,0.25,and 0.19.In vitro,the expression of hUAT mRNA in HK-2 cells is associated with the concentration of sexual hormones.
