Abstract

Objective To evaluate the effect of sevoflurane anesthesia on trafficking of GluR2-containing α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptors in aged rat hippocampi.Methods Sixty-eight healthy male Wistar rats,aged 18-20 months,weighing 600-650 g,were randomly divided into 2 groups (n =34 each) using a random number table:control group (group C) and sevoflurane group (group S).In group S,open tibial fracture was performed and anesthesia was induced with 3.6% sevoflurane for 2 h The contextual fear conditioning test,tone-cued fear conditioning text and Y maze test were performed on days 1,3 and 7 after surgery and the expression of GluR2 subunit-containing AMPA receptors in total and membrane proteins in hippocampi was detected by Western blot.The ratio of AMPA receptors in the membrane protein to those in the total protein (m/t ratio) was calculated.Results Compared with group C,the percentage of freezing time in the contextual fear conditioning text and percentage of spontaneous alternation were decreased,the expression of GluR2 subunit-containing AMPA receptors in the membrane protein was down-regulated,the m/t ratio was decreased on days 1 and 3 after surgery,and the percentage of freezing time in the tone-cued fear conditioning text was decreased on days 1-7 after surgery in S group (P < 0.05).Conclusion The mechanism by which sevoflurane anesthesia induces the impairment of learning and memory function in aged rats is related to enhanced trafficking of GluR2-containg AMPA receptors from cell membrane to cytoplasm in hippocampi.
