Abstract

肝癌是全世界范围的高发恶性肿瘤,在亚洲的发病率、病死率都很高。肝癌的分子学发病机制尚不完全清楚,包括基因结构和功能的缺陷、癌基因的激活、抑癌基因的失活、细胞周期紊乱、染色体的非整倍体出现以及细胞凋亡途径受阻等。垂体瘤转化基因(PTTG)1是细胞周期调节蛋白,能抑制姐妹染色单体分离,导致染色体不稳定,形成非整倍体。在成人大多数正常组织中PTTG1表达较弱,甚至检测不到,而在垂体肿瘤等多种肿瘤细胞中表达明显升高,PTTG1基因可能是一种潜在的抗癌基因治疗靶点[1]。本研究用小干扰RNA (siRNA)降低肝癌细胞株HepG2、SMMC-7721细胞中PTTG1的表达,检测PTTG1表达下调对肝癌细胞增殖及凋亡的影响,探讨PTTG1对肝癌细胞生物学行为的影响以及作为肝癌治疗靶基因的可能性。 关键词:癌,肝细胞; 垂体瘤转化基因1;RNA干扰
