Abstract

Objective To observe autophagy of prostate cancer PC-3 cells induced by resvera-trol and explore the possible mechanism.Methods Prostate cancer cell line PC-3 was incubated in culture medium in vitro,using cell counting kit-8 assay to detect the growth rate.Reverse transcriptase-polymerase chain reaction (RT-PCR) method was used to examine the expression level of LC3 and Becline 1.Weastern blotting method was used to examine the expression level of LC3-Ⅱ.Results After oridonin treatment,the proliferation of PC-3 cells was inhibited significantly in a concentration and time-dependent manner.With the concentration of 20 μmol/L and 24 h,the inhibition ratio was (9.21 ± 1.10)%,when the concentration wan 160 μmol/L and 72 h,the inhibition ratio grow up to (65.73 ± 0.69) %.There was different elevation of the LC3,Beclin 1 mRNA expression in different concentration groups of PC-3 cells after being treated resveratrol.After treatment with different concentration of resveratrol for 24 h,the levels of LC3-Ⅱ proteins expression were up.Conclusion We comfirmed that resveratrol had the capability of inhibiting the proliferation of PC-3 cells in vitro by a dose and time-dependent manner.Resveratrol may induce autophagy leading cells death to inhibit proliferation of prostate cancer cells.
