Abstract

Neutrase AS1.398 and alkalase 2790 were added into AOT/ iso-octane reverse micelles for separating soybean protein and oil from soybean powder. The composition of protein extracted were examined. Gamma component of soybean protein showed differences between reverse micelles extraction and the traditional water extraction technology showed by Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Low-molecular-weight components increased when protease was added in the reverse micelles and new components appeared about 5 hours of enzyme action. The effect of neutrase AS1.398 was more significant that that of alkalase 2709. With 2hr extraction, the yield of low-molecular-weight components increased. For 5hr extraction, new components accounted for 21.17% and 7.76% for AS1.398 and alkalase 2709 respectively. Scanning electron microscopy testified that enzymatic protein granule was much finer, which conformed to the outcome of electrophoresis. It was concluded that enzymatic hydrolyzed protein and oil can be separated simultaneously through reverse micelles with proteases added.
