Abstract

Objective To observe the effect of microRNA-19b(miRNA-19b) on necrosis of pancreatic acinar cells in acute necrotic pancreatitis(ANP). Methods The model of ANP was established by adding the aurolithocholic acid 3-sulfate disodium salt(TLC-S)of 200μmol/L into the rat pancreatic acinar cell line(AR42J), 0. 5 ml every well,and blood amylase was determined after 1 h.The real-time reverse transcriptase-polymerase chain reaction(RT-qPCR) was used to detect the expression of miRNA-19b.The plasmid carrying the miRNA mimics and antisense oligodeoxynucleotide of miRNA-19b was transfected into the AR42J cells.The control group was transfected with vehicle control.The acridine orange)/ethidium bromide(AO/EB) reagent was used to detect the necrosis of acinar cells. Results The expression of miRNA-19b in the experimental group was increased by(2. 51±0. 14) times as compared with the control group(P< 0. 05). The expression level of miRNA-19b in the AR42J cells transfected with mimics was elevated by(5. 94±0. 95) times as compared with the vehicle groups(P< 0. 05).The necrosis of acinar cells was aggravated in the miRNA mimics-transfected group as compared with the vehicle groups[(50. 3±1. 5)% vs. (39. 6±2. 3)%, P< 0. 05]. Conclusion The expression of miRNA-19b was increased in ANP. The necrosis was aggravated by up-regulating the expression of miRNA-19b, and alleviated by down-regulating the expression of miRNA-19b.
