Abstract

Objective To investigate the effect of mesoderm specific transcript (MEST) expression on the migration and proliferation of rat osteoblasts.Methods siRNA interference fragments were used to knock down the MEST expression in osteoblasts.Transwell assay was used to detect the migration ability,and cell counting kit-8 (CCK-8) and the detection of proliferation cell nuclear antigen (Ki-67) proliferation index were used to assess the proliferation ability.Results After MEST down-regulation,the number of migratory osteoblasts was (10.0 ± 1.9),significantly less than the mock-vehicle group (23.5 ±2.1) and control group (25.2 ± 1.8),with a statistically significant difference (P < 0.05).Meanwhile,CCK-8 assay revealed that MEST down-regulation significantly reduced the proliferation ability of osteoblasts,and Ki-67 expression was also significantly decreased.Conclusion MEST expression can promote the migration and proliferation of osteoblasts.
