PROCEDURE

Mitochondria were isolated from livers of rabbits that had been fasted for 24 hours. Sucrose, 0.25 M buffered with Tris-. HCl pH 7.4, 0.0025 M, was used throughout as the suspending medium during homogenization, incubation, and washing. As the standard experimental procedure, aliquots of twice washed, freshly prepared mitochondria were incubated for 15 minutes in the presence of 0.3 PC of K& and the mercurial agent under investigation. The incubation media contained reagents required for oxidative phosphorylation. These are listed in the legend for Fig. 1. The incubations were terminated by centrifugation at 8000 X g for 5 minutes, after which the precipitate was washed twice. 1 The abbreviations used are: EDTA, ethylenediaminetetraacetic acid, and CMB, p-chloromercuribenzoate.
