Abstract

Objective To detect the expression of long noncoding RNA (lncRNA) TCONS_00023297 in osteosarcoma cells and its effect on the proliferation and migration of osteosarcoma cells. Methods The expression of lncRNA TCONS_00023297 in osteosarcoma cell lines was detected by real-time fluorescence quantitative PCR (FQ-PCR). A cell line with high and low expression of lncRNA TCONS_00023297 was constructed. The effects of high and low expression of lncRNA TCONS_00023297 on migration and invasion of osteosarcoma cell line U2OS were detected by cell scratch and Transwell assay. The effects of high and low expression of TCONS_00023297 on the proliferation of osteosarcoma cell line U2OS were detected by cell counting kit-8 (CCK-8) and clonogenic assay. Western blotting and FQ-PCR were used to detect the effects of high and low expression of lncRNA TCONS_00023297 on the expression of proliferation-related genes CDK4 and p21 in osteosarcoma cell line U2OS. Results FQ-PCR results showed that the expression of lncRNA TCONS_00023297 in U2OS cells (11.02±0.35) was significantly higher than that in human bone marrow mesenchymal stem cells (hBMSCs) (1.00±0.11) (t=61.070, P<0.01). Scratch test showed that the mobility of lncRNA TCONS_00023297 overexpression group [(87.12±3.98)%] was significantly higher than that of control group [(65.47±5.69)%] (t=6.971, P<0.05). Transwell assay showed that the number of invasive cells in the overexpression group of lncRNA TCONS_00023297 (198.5±22.36) was significantly greater than that in the overexpression control group (128.50±16.25) (t=5.662, P<0.05). CCK-8 and clonogenic assay showed that the proliferation of U2OS cells was enhanced in the overexpression group of lncRNA TCONS_00023297 as compared with the control group. As compared with the control group, the proliferation of U2OS cells decreased in the low expression group of lncRNA TCONS_00023297. Western blotting and FQ-PCR results showed that the expression of CDK4 in U2OS cells increased, and that of p21 decreased in the lncRNA TCONS_00023297 overexpression group as compared with the control group. As compared with the control group, the expression of CDK4 in U2OS cells decreased and that of p21 increased in the low expression group of lncRNA TCONS_00023297. Conclusion Overexpression of lncRNA TCONS_00023297 promoted U2OS cell proliferation, migration and invasion. Key words: Long noncoding RNA; Osteosarcoma; Proliferation; Migration
