Abstract

Objective To investigate the effect of juanyin xiefei formula on lipopolysaccharide (LPS) induced the expression of mucin MUC5AC and explore the possible mechanism of regulation. Methods Human bronchial epithelial cells 16HBE were cultured in vitro.The cells were divided into 0, 0.5, 1, 6, 12 and 24h groups according to the length of LPS stimulation, and the LPS stimulation length of follow-up experiments was determined.The CCK8 method was used to detect the ability of juanyin xiefei formula about different concentration on cell proliferation of 16HBE cells and cells after stimulation with LPS, the optimum concentration of drug was selected.The experiment was divided into control group, LPS stimulation group and juanyin xiefei formula Group, using Western blot method on detection of MUC5AC and suppressor of cytokine signaling 1(SOCS1) protein expression. Results The high expression of MUC5AC protein induced by LPS, 0, 0.5, 1, 6, 12 and 24 h groups, the relative expression of MUC5AC gradually increased, while the relative expression of SOCS1 protein gradually decreased.The expressions of MUC5AC and SOCS1 were both medium up-regulate in 6 h group.Consequently, the application of LPS for 6 h was selected as the optimal responses period in the ensuing experiments.According to the experimental results of CCK-8 when cultured in high concentration juanyin xiefeiformulahas a growth inhibiting effect on 16HBE, when the concentration of 0.005% juanyin xiefei formula on the treatment effect of cells after LPS stimulation was the best.The concentration of follow-up drugs was 0.005%, and the action time was 48 h. After LPS treatment, the relative expression of MUC5AC in 16HBE cells was 0.251 3±0.004 3, LPS could stimulate MUC5AC expression, and the relative expression of SOCS1 was 0.427 9±0.004 0, SOCS1 expression was inhibited.After the treatment of juanyin xiefei formula and LPS, the relative expression of MUC5AC was 0.131 8±0.002 9, and decreased by 0.144 8±0.004 7.Juanyin xiefei formula inhibited the expression of MUC5AC induced by LPS (P<0.01). The relative expression of SOCS1 was 0.572 7±0.008 6, and increased by 0.119 4±0.002 3.Juanyin xiefei formula promoted the expression of SOCS1 after the action of LPS (P<0.01). Conclusions Juanyin xiefei formula could inhibit the expression of LPS induced airway mucin MUC5AC, the mechanism may be related with SOCS1.
