Abstract

Objective To investigate the effect of ischemia and reperfusion on expression of Hedgehog signaling in the liver after rat partial hepatectomy.Methods The model of rat partial hepatectomy was established by resecting the middle and left lobes of the liver.Eighteen Sprague-Dawley rats were randomly divided into 3 groups:sham operation (A),oartial hepatectomy (B),partial hepatectomy combined with hepatic ischemia and reperfusion (C).Blood and hepatic tissues were collected 48 h after the operation Plasma alanine transaminase (ALT) and the expression of proliferation cell nuclear antigen ( Ki-67 ),Sonic hedgehog ( Shh),Indian hedgehog (Ihh) and glioma-assocated oncogene-2 (Gli-2) in the hepatic tissue were detected.Results As compared with group A,the plasma ALT was increased in groups C (P < 0.01 ) and B (P >0.05 ).As compared with group B,the plasma ALT was increased in group C (P<0.01 ).Adipose degeneration and cells at different division stages were shown in hepatic tissue of groups B and C.As compared with group A,the expression of Ki-67,Shh,Ihh and Gli-2 in groups B and C was increased significantly ( P < 0.01 ).and that was higher in group C than in group B ( P < 0.05 ).Conclusion Hedgehog signaling may be activated after rat partial hepatectomy,which can promote the liver regeneration.Ischemia and reperfusion may enhance the activation of Hh signaling after partial hepatectomy.
