Abstract

目的 探讨缺氧环境对人肝癌细胞侵袭能力的影响.方法 用Transwell小室筛选高侵袭能力的HepG2细胞株,用氯化钴(150μg/ml)对该高侵袭细胞株进行化学诱导缺氧培养,分别于培养24,48,72 h后,用Western印迹法检测HIF-1α蛋白的表达,用RT-PCR方法检测CD44v6mRNA的相对表达量,统计学分析两者的改变.结果 随着缺氧培养时间的增加,HIF-1α蛋白和CD44v6 mRNA的表达量的改变具有显著性差异,并且两者呈同步改变.结论 缺氧可以通过增加肝癌细胞的迁徙能力促进肿瘤的转移。
