Abstract

Objective To investigate the effects of reactive homocysteine (Hcy) on the mitochondrial membrane potential (JC-1) and reactive oxygen species (ROS) production in human umbilical vein endothelial cell (HUVEC). Methods HUVEC were isolated and cultured , treated with Hcy in different concentration(0mmol/L,0.01 mmol/L,0. 1 mmol/L,1 mmol/L,3 mmol/L,5 mmol/L) and for different duration(3 h,6 h,12 h,24h) , JC-1 and ROS were detected with the flow cytometry. Results The JC-1 decrease(F = 101. 82,F = 85.257,P<0.01) and ROS increase (F = 89. 82,F = 82. 235,P <0. 01), had a significant difference.Conclusion Hcy could induce HUVEC injury by decreasing MMP and producing ROS in a time- and dose-dependent manner.
