Abstract

Objective To observe the effect of fumigation No.1 on rat wound repair after operation of anal fistula in angiogenesis and microcirculation and discuss its mechanism. Methods Selection of 60 male SD rats were used for postoperative anal fistula model. After the success of modeling, the SD rats were divided for four groups: fumigation No.1 group, potassium permanganate group, physiological saline group and blank control group, 15 rats in each group. At first, 7 and 14 days, 5 rats were randomly selected from each group to test the following indicators: The wound blood flow, the expression of v-ascular endothelial growth factor (VEGF) and CD34 count from Granulation tissue of wound by Immunohistochemical method. Results Fumigation No.1 can improve the expression of VEGF and CD34 count in the early and middle period of wound healing, wound increase local blood flow. Began to increase in first days [local blood flow, VEGF, CD34 respectively as (69.29±2.29) ml/min, 0.34±0.02, 0.50±0.03; the same below] after surgery, seventh days [(212.22±1.87) ml/min, 0.70±0.03, 0.66±0.03] to reach the peak, and then began to decline. In the later stage of wound repair(Fourteenth days), in the fumigation group [(47.73±2.48) ml/min, 0.89±0.03, 0.79±0.03] and potassium permanganate group [(69.86±1.63) ml/min, 0.63±0.03, 0.68±0.05]. The wound blood flow Reduced, the expression of VEGF and CD34 count from Granulation tissue of wound decreased. In the treatment of 7 d and 14 d, the difference was statistically significant (P=0.016) between fumigation group and normal saline group [(100.28±1.67) ml/min, 0.44±0.03, 0.51±0.02] [(85.57±1.83) ml/min, 0.52±0.03, 0.60±0.03], blank control group [(98.78±1.32) ml/min, 0.21±0.03, 0.39±0.03] [(82.36±1.87) ml/min, 0.22±0.04, 0.38±0.03] when it comes to the wound blood flow, the expression of VEGF and CD34 count from Granulation tissue of wound. In the treatment of 7 d, the difference that The wound blood flow, the expression of VEGF and CD34 count from granulation tissue of wound was also statistically significant (P=0.025) between fumigation group (Significantly elevated) and potassium permanganate group [(153.43±1.46) ml/min, 0.56±0.02, 0.58±0.03]. In the treatment of 14 d, the difference of those indicators except CD34 count was also statistically decreased significantly (P=0.017). Conclusion Fumigation No.1 has good regulative effects on Local wound blood flow, the expression of VEGF and CD34 count from granulation tissue of wound.
