Abstract
Purpose: Production of endothelin (ET-1) increases under pathophysiological conditions such as heart failure. ETA receptor is one of the Gq-coupled receptors and we have previously reported that stimulation of alpha1-adrenoceptor, another Gq-coupled receptor, increases L-type Ca current (ICa) via activation of protein kinase C (PKC) and Ca/calmodulin dependent protein kinase II (CaMKII) using a perforated patch clamp technique (O-Uchi et al, PNAS 2005;102:9400-5). Thus, ETA receptor stimulation was expected to show a similar response of ICa to that in the case of alpa1-adrenoceptor stimulation.
