Abstract

Objective To explore the effect of Danggui Buxue decoction on angiogenesis, Ang-1, Ang-2, Tie-2, and VEGF in rats with cerebral ischemia reperfusion. Methods Rat models of the middle cerebral artery occlusion/reperfusion (MCAO) were created using suture method. The rats models were randomly divided into a sham operatio group, a model group, a low-dose group, and a high-dose group. Each group was continuously treated with correspondent drugs for 14 days. Neurological function 1 and 14 days after operation was evaluated with Garcia nerve function score standard. The microvessel density and Tie-2 expressions in the ischemic area and the surrounding area were evaluated by immuno histochemistry. The VEGF expressions in ischemic area and the surrounding area were evaluated by Western Blotting. Results The nerve function scores, the concentrations of MVD, Ang-1, and Ang-2, the levels of Tie-2 and VEGF after continuous administration for 14 days were significantly higher in the low-dose group and the high-dose group than in the model group; and the indicators significantly increased with Danggui Buxue decoction dose. Conclusions Danggui Buxue Decoction can promote the recovery of nerve function in rats with cerebral ischemia reperfusion injury; the mechanism is possible that promoting angiogenesis in cerebral ischemic area increasing the expressions of Ang-1, Ang-2, Tie-2, and VEGF.
