Abstract

Objective To detect the effect of cellular prionprotein (PrPC) expression on proliferation and invasion of colorectal cancer cells.Methods PrPC expression was detected in HCT-116,SW480 and Lovo cell lines by real-time reverse transcriptase-polymerase chain reaction (RT-qPCR) and Western blot.After transfecting the Lovo and SW480 cell lines by PrPC spefical lentivirus vectors,the ability of proliferation and invasion was evaluated by methyl thiazol tetrazolium (MTT) assay and Transwell assay,respectively.Results The mRNA level of PrPC was higher in Lovo and SW480 cell lines (8.19 ± 1.12,7.48 ± 1.21,respectively) compared with those in HCT-116 cell line (3.56 ± 1.03).After downregulation of PrPC,cell proliferation and invasion were significantly inhibited in Lovo and SW480 cell lines,with a significant difference (P < 0.05).Conclusion PrPC expression is closely associated with the proliferation and invasion of colorectal cancer cells.
