Abstract

Objective To explore the effect of angiotensin Ⅱ on F-actin in podocytes and the protective role of AT1 receptor antagonist for F-actin. Methods Mouse podocytes were stimulated by angiotensin Ⅱ of 10-6 mol/L for 10,30,60,and 120min,then were devided into five groups: control,angiotensin Ⅱ of 10-10,10-8,and 10-6 mol/L,and losartan of 10-6 mol/L plus angiotensin Ⅱ of 10-6 mol/L. The expressions of F-actin were detected. Results Angiotensin Ⅱ decreased the expression of F-actin in a time- and concentration-dependent manner. Angiotensin Ⅱ destroyed the morphology of F-actin,making it from orderly arrangement to loose structure. Losartan blocked the effect of angiotensin Ⅱ on F-actin. Conclusions Angiotensin Ⅱ can destroy F-actin of podocytes by AT1 receptors,and AT1 receptor antagonist can protect podocytes. Key words: Angiotensin Ⅱ; Losartan; Podocytes; F-actin
