Abstract

Aminophylline inhibited platelet aggregation in human platelet-rich plasma. This inhibition was reversed by adrenaline at concentrations of adrenaline insufficient to induce platelet aggregation. The action of adrenaline was inhibited by phentolamine but not by propranolol. Aminophylline had no effect on the release of adenine nucleotides or the breakdown of radioactively labelled ATP induced by thrombin in washed, isolated platelets. These results suggest that the mechanism of inhibition of platelet aggregation by aminophylline is unconnected with the platelet “release reaction”. The observed action of adrenaline suggests that agonists acting at alpha-adrenergic loci may lead to reduction in platelet cyclic AMP.
