e15167 Background: Presence of cancer stem cells (CSCs) in hepatocellular carcinoma (HCC) are highly potential to evade current therapeutic regimen and responsible for tumor recurrence. Studies reported that expression of dysadherin in CSCs could be used as a prognostic indicator, also responsible for treatment failure and poor survival rates. In the current study, we analyzed the role of enhanced dysadherin in drug and apoptosis resistance in human HCC SP cells. Methods: SP cells from 19 human HCC patients were sorted by FACs. RNA interference evaluated the expression of stemness genes(CD133, ABCG2, OCT-4, EpCAM, BCL-2, BAX) in SP cells before and after dysadherin knock down using RT-PCR analysis. Immunohistochemistry (IHC) assays assessed dysadherin protein expression in SP cells.
