Abstract

Objective:To investigate the effect of 2,3,4′,5-tetrahydroxystilbene-2-O-β-D-glucoside(TSG)on the expression of insulin-like growth factor-1(IGF-1)in myocardial tissue of diabetes rats.Methods:Diabetes model rats was developed by feeding a high-fat diet and single intraperitoneal injection of 1% streptozocin(STZ,30mg/kg).Forty STZ-diabetic rats were randomly divided into 4groups:the diabetes model,the low-,middle-and high-dose TSG groups.In the low-,middle-and high-dose groups,rats were administered TSG 30,60 and 120mg/kg by gavage once daily for 6 weeks.Another 10 rats were used as controls.Body weight,food and water consumptions were collected and fasting blood glucose(FBG)was determined.Enzyme-linked immunosorbent assay(ELISA)was used to analyze the content of serum IGF-1 and immunofluorescence was applied to determine the expression of IGF-1 in myocardial tissue.Results:The content of serum IGF-1 and the expression of myocardial IGF-1 in diabetes model rats were decreased(P0.05,P0.01)when compared with the controls. The content of serum IGF-1 and the expression of myocardial IGF-1 in the middle-and high-dose TSG groups were increased(P0.05,P0.01)when compared with diabetes model rats.Conclusion:TSG could increase serum IGF-1 level,up-regulate the expression of myocardial IGF-1.
