Abstract

Background: Bone lead is a significant predictor of decline in scores on the Mini Mental Status Exam (MMSE) in older adults, indicating that accumulated lead burden negatively impacts cognitive function. It is unknown if genes associated with Alzheimer's disease and cognitive impairment modify the association between lead and cognitive function. Aims: To evaluate whether APOE (rs429358 and rs7412), PICALM (rs3851179), CLU (rs11136000) CR1 (rs6656401), and CELF1 (rs7933019) polymorphisms found to be associated with Alzheimer's disease in genome-wide association studies modify the association between cumulative lead exposure and incident cognitive impairment defined by having an MMSE score
