Abstract

Objective To investigate the effect and significance of cinobufacine on the expression of survivin in the ovarian carcinoma 3AO cells cultured in vitro. Methods Ovarian carcinoma 3AO cells were cultured in vitro Following intervention of cinobufacine with different concentrations, the expression of survivin was measured by RT-PCR, and cell apoptosis was tested by flow cytometry. Results For the control group, the expression of survivin mRNA in the ovarian carcinoma 3AO cells was 1. 01 + 0. 13, cell apoptotic rate was (2. 31 ± 0.98)%. Nevertheless, 0. 25 mg/ml cinobufacine didn't affect survivin and cellular apoptotic rate (P > 0. 05 ). Following administration of 0.25 mg/ml cinobufacine, the expression of survivin mRNA was 0.67 ± 0.16, and cell apoptotic rate was (28.69 ± 4. 16)%. Statistical differences could be seen when it was compared with that of ovarian carcinoma 3AO ceils in the control group( P < 0.05 ). There were no statistical differences in the effect that survivin mRNA expression and cell apoptotic rate were affected following applications of either 25 mg/ml or 2.5 mg cinobufacine ( P > 0.05 ). Conclusions 2.5 mg/ml cinobufacine could inhibit the expression of ovarian carcinoma 3AO cells. Key words: Cinobufacine; Ovarian carcinoma; 3AO cell; Survivin; Apoptosis
