Abstract

INFECTION of animal cells with picornaviruses results in inhibition of host cell RNA and protein synthesis shortly after infection, as was shown with members of the cardiovirus group1–3, with poliovirus4–6, and with foot-and-mouth disease virus7. The molecular mechanism of this so-called shutoff is not well understood; however, it seems to require protein synthesis early in infection2,8,9, but no viral RNA replication9–11. We report here that 2-thio-4-oxothiazolidine (rhodanine), at a concentration which inhibits selectively the uncoating of echovirus 12 in GMK cells also inhibits the virus-induced shutoff of GMK cell protein synthesis.
