Abstract

The aim of this study was to evaluate the effect of administration of a high fat diet after weaning, during 8 weeks, in renal morphology and metabolic profile. Weaned male Wistar rats (21 days, n=8/group) were randomized divided in Control group (CG), commercial rat feeding, and Hyperlipidic group (HG) by addition of 30% unsalted butter in commercial diet to rats during 8 weeks. The left kidney was collected under anesthesia (sodium pentobarbital, 50mg/kg) to histological glomerular analysis (5μm thick, Picrosirius Red) to evaluate Glomerular Tuft Area (GTA) and Bowman's Capsule Area(BCA). Blood was collected to biochemical assays: basal glycemia, triglycerides and total cholesterol using the enzymatic colorimetric kits (Labtest, BR). There was no change in basal glycemia (CG: 92 ± 4 vs HG: 85±4 mg/dL). However, HG showed an increase in Triglycerides (CG: 51 ±5 vs HG: 78±6 mg/dL) and Total Cholesterol (CG: 75 ±4 vs HG: 101±10 mg/dL). There was an important reduction in GTA (26%) and in the BCA (22%) in HG group. At the end of the protocol, we did not find differences in bodyweight between the groups, however, the adiposity was increased in HG (15 ±1g) when compared with CG (5.2±0.5 g). These data suggest high fat diet during lifespan induced metabolic alterations associated with earlier kidney injury. Supported By: Mackpesquisa (059/2011).
