Abstract

Purpose: Physical activity is recommended for the general management of symptoms in OA patients. Despite this fact, mechanical loading is rarely a feature implemented in early drug screening tools. Pre-clinical and clinical studies have shown beneficial effects of loading on cartilage extracellular matrix (ECM) turnover and moderate exercise in joint mobility, respectively. Applying mechanical load to cartilage in ex vivo models may improve in vivo “likeness” and aid translation by promoting increased understanding of how the therapeutic potential of mechanical loading may interact with disease-modifying osteoarthritis drugs (DMOADs) in development.
