Abstract

Abstract A special double-cladding optical fiber with three-layer step-index structure was introduced for simplifying the stimulated-emission-depletion (STED) microscopy. With this special fiber, a dark hollow beam with decent doughnut-shaped focal spot can be generated for incident light in a specific wavelength range. At the same time, a Gaussian-like beam can be generated when the wavelength of incident light is out of (either longer or shorter than) the specific range. An all-fiber STED illumination system has been achieved by this special fiber. For special cases, a semi-fiber solution based on this special fiber has also been realized.
