Abstract

Objective To observe the chimerism in small bowel transplantation (SBT) rat mod-el. Methods 15 Male BN and 15 female Lewis rats were used as donors and recipients, which were as-signed randomly to three groups, control group, FK506 group, PV group (FK506 + infusion donor bone marrow cells via portal vein) ;Fluorescence in Situ Hybridization and realtime polymerase chain reaction (PCR) for sex-determining region of Y chromosome(SRY) was used to determine the location and con-centration of male cells in recipients' various tissues, including blood, liver, spleen. Results The forma-tion of donor-derived chimerism in the bone marrow cells infusion groups [blood (3.03±0.16)% ,liver (0.86 ± 0.05)%, spleen (4.08 ± 0, 12)%] was statistically higher than FK506 group [(1. 15 ±0.07) %, (0.21 ± 0.02) %, (1.23 ± 0.05) %] and control groups [(1. 10 ± 0.07) %, (0. 22 ±0.02) %, (1.17±0.06) % ,P <0.01]. Conclusion Mix chimerism status were established by infusion of donor-derived bone marrow cells. Key words: Chimerism; Rat; Small bowel transplantation; Bone marrow cells
