Abstract

Twenty solvent systems were tested to determine whether they are applicable to separation of DNP-amino acids by silica gel thin-layer chromatography. It was found that among tested solvent systems, isoamyl alcohol saturated with 5% acetic acid (I), n-butanol saturated with water (II) and chloroform : benzyl alcohol : acetic acid (70:30:3) (III) were the most useful for the chromatography. Excellent two-dimensional chromatograms were obtained, using solvent I followed by solvent II or III. The possibility of quantitative determination of DNP-amino acids was also investigated.
