Abstract

Thin-layer chromatography of α-keto acids A method is described for the qualitative and quantitative estimation of α-keto acids as their 2,4-dinitrophenylhydrazones by thin-layer chromatography on silica gel. The solvent system is composed of ethyl formate, petroleum ether, and glacial acetic acid. The method id suitable for the separation of six keto acids of biological interest, viz. oxaloacetic acid, α-ketoglutaric acid, glyoxylic acid, pyruvic acid, α-ketobutyric acid, and α-ketoisovaleric acid. The quantitative assay is carried out spectrophotometrically at 380 nm after elution of the separated substances from the thin-layer plates.
