Abstract

Objective To investigate the distribution of HBV genotypes and subgenotypes in patients with HBV related hepatocellular carcinoma (HCC) in Lanzhou,and discuss the relationship between the genotypes/subgenotypes of HBV and HCC.Methods The patients were classified into 3 clinical groups:chronic liver disease patients (CHB group),liver cirrhosis patients (LC group) and HCC patients (HCC group).A total of 142 serum samples were collected from these patients,including 62 HBeAg positiye samples and 80 HBeAg negative samples.HBV DNA of the samples were extracted and detected for the mutation in S gene.The pre-S and S gene sequencing were introduced to NCBI online genotyping to determine HBV genotypes.The phylogenetic trees were constructed by MEGA4 to analyze HBV subgenotypes.Results There were 19 samples with genotype C(9 samples with C1,10 samples with C2),17 samples with genotype B(all with B2),2 samples with B/C,and 6 samples with genotype D(all with D2) in CHB patients.In LC group,there were 26 samples with genotype C(9 as C1 and 17 as C2),8 samples with B2 and 2 samples with D2.In HCC group,there were 23 samples with genotype C(5 as C1,18 as C2),6 samples with B2 and 1 sample with B/C,2 samples with D2.There were significant differences of subgenotype C2 in three groups(x2 =10.498,P < 0.05).Conclusions The genotypes in Lanzhou chronic HBV infection are genotype B(B2),C (C1,C2),D(D2) and B/C.The genotype C detection rate in HCC group and LC group are obviously higher than CHB group.The genotype C related to serious liver diseases,such as LC and HCC.
