DNA·RNA hybrids are non-canonical nucleic acid structures of fundamental importance for gene regulation and disease. In this issue of Chem, Terrazas et al. investigate the origins and biological consequences of the DNA·RNA asymmetry and deliver novel insights for improving applications of key enzymes, such as RNase H and CRISPR-Cas9.
