Abstract

5α-Dihydrotestosterone has been shown to be a potent androgenic hormone comparable with testosterone and to be an active form of testosterone in target organs in rats (Gloyna & Wilson, 1969; Tveter & Aakvaag, 1969), and in stallions and bulls (Jeffcoate & Short, 1970). In the domestic fowl the conversion of testosterone to 5α-dihydrotestosterone by the coccygeal gland and comb has been reported by Gloyna & Wilson (1969). The present study was conducted to elucidate the conversion of testosterone to 5α-dihydrotestosterone by the epididymis of the domestic fowl. Epididymides removed from 10-month-old White Leghorn cocks were homogenized in ice-cold 0·25 m-sucrose solution with a loose-fitting glass-Teflon homogenizer. The homogenate (2 ml) was incubated for 60 min in 100% oxygen with [4-14C]androstenedione or [4-14C]testosterone in 2 ml 0·25 m-sucrose solution containing NADPH at a final concentration of 120 μmol/l.
