Abstract

Misonidazole, a 2′nitroimidazol, has a high electron affinity and produces in cell culture and on tumors in experimental animals appreciable sensitization for hypoxic cells. We conducted a long term study using the regrowth delay assay for the response of single and fractionated x-ray treatments and in combination with misonodazole testing two serially xenografted human squamous cell carcinomas of the head and neck region. The greatest improvement is for single dose treatment with a misonidazole-enhancement-ratio of 1.8 because in this case hypoxic cells are a big limitation in as much there is no opportunity for reoxygenation. There is no gain due to the sensitizer for the fractionation pattern of 5 fractions weekly.
