Abstract
PBMCs of patients undergoing haemodialysis are at significant risk to undergo AICD. To investigate if plastic tubings, as utilized in DIALYSIS, might cause defects in blastogenesis assays (CD3 mAB, 10 μg) or induce a state of susceptibility of leukocytes to undergo AICD. A MOCK dialysis circulation was set up and medium was drawn after 120 minutes. 1:4 diluted medium inhibited proliferation in blastogenesis assays (thymidine incorporation) by a mean of 58,8±8.4%(p<0.001) relative to control. After incubation with SN vs medium for 24 hours and/or LPS (10ug/cc) and consecutive coculture with antiCD3/28 (10ug/cc) mABs for 18 hours, AICD, as measured by CD4/7-AAD positivity by FACS analysis, increased by 4.07 (p<0.001), 10.22 (p<0.0001), respectively. Since a inorganic soluble substance deemed to induce immunesuppression, GCMS was utilized and DEHP at a concentration of a mean 3.3 ppm was detected. A dosedependent immunesuppression was verifyed in a raw substance proliferation assay (p<0.001). These results indicate that leaching plastic softeners are inducing immunesuppression.
