Abstract

Objective To study the effect of dexamethasone on the expression of aquaporin 1 (AQP1)in pleural mesothelial cell patients with pleural fluid. Methods Human pleural mesothelial cell was obtained from the patients with benign and non-infective pleural fluid and cultivated after preconditioning the first generation cell by 0μmol/L, 1 μmol/L, 100μmol/L dexamethasone respectively,and the expression level of AQP1 protein and mRNA was detected by western blotting and RT-PCR separately. Results The survival rate and purity of human pleural mesothelial cell isolated and cultivated from pleural fluid are both above 95 %,the expression level of AQP1 protein and mRNA of human pleural mesothelial cell induced by dexamethasone are raised up significantly. The expression level has direct correlation with the density of dexamethasone. Conclusions Dexamethasone can up-regulate AQP1 gene expression dose-dependent at translational and transcriptional level. And then accelerate the absorption of hydrothorax.
