Abstract

The protein crystallography beamline (PX-BL 21) at Indus-2 synchrotron (RRCAT, Indore) is dedicated for structure determination of macromolecules. For solving the phase problem in structure determination of macromolecules, MAD (Multi-Wavelength Anomalous Diffraction) scan is one of the well established methods. This scan requires a precise measurement of absorption data at or near the absorption edge of doped heavy atom for different wavelengths of X-rays. Software for absorption edge scanning using fluorescence detector and double crystal monochromator has been developed and deployed at PX-BL21. MAD experiment of a protein sample has been recorded and analyzed. With this important development, PX-BL21 is now fully equipped to perform anomalous diffraction experiments with single crystal protein samples.
