Abstract

ABSTRACT The medicinal properties and toxicity of aconitine in traditional Chinese medicine have caused widespread concern. In this study, we screened a highly specific and sensitive monoclonal antibody with an IC50 of 3.65 ng/mL generated by immunizing mice and cell fusion, and the limits of detection of the ic-ELISA were 1.13 and 11.76 ng/mL. We then developed a colloidal gold test strip based on the antibody. For rapid detection of aconitine in herbs, the visual limit and detection limit of the test strip are 10 and 25 ng/mL, respectively; the results are obtained within 5 min and are visible to the naked eye. Therefore, the colloidal gold test strip is very suitable for on-site detection and quality screening of aconitine in Chinese herbal medicine.
