Abstract

A selective and sensitive liquid chromatographic/tandem mass spectrometric method was developed and validated for the matrix effect controlled estimation of sildenafil and N-desmethyl sildenafil in human plasma. The samples were extracted on solid-phase extraction cartridges and chromatographed on XTerra MS C18 5 µm (4.6 mm x 150 mm) column using isocratic mobile phase. The method was validated over the concentration range of 5.00–1401.35 ng/mL for sildenafil and 2.83–603.38 ng/mL for N-desmethyl sildenafil. The parent to product ion transitions for sildenafil, N-desmethyl sildenafil and quetiapine were m/z 475.10–283.30; 461.20–283.20 and 384.20–253.20, respectively. Matrix factor was assessed using post-extraction addition technique and found to be controlled.
