Abstract

A simple, precise, rapid, and accurate high-performance thin-layer chromatographic (HPTLC) method has been developed for the simultaneous estimation of thiocolchicoside (THIO) and aceclofenac (ACE) in bulk and pharmaceutical dosage form on silica gel precoated aluminum 60F254 plates (10 cm × 10 cm) with 250 mm thickness. The separation was carried out in methanol-chloroform-water 9.6:0.2:0.2 (v/v/v) as mobile phase. The densitometric scanning was carried out at 254 nm. The RF values were found to be 0.70 ± 0.05 for THIO and 0.83 ± 0.05 for ACE. The linearity was obtained in the range 30–180 and 750–4500 ng per band for THIO and ACE with correlation coefficients r2 = 0.9995 and 0.9993, respectively. The method was validated for specificity, linearity, accuracy, precision, limit of detection (LOD), limit of quantification (LOQ) and robustness.
