Abstract

Primer pairs for ten microsatellite loci of the Wolfie Cardinalfish, Cheilodipterus artus (Apogonidae) were designed from a partial genomic DNA library. The loci were optimised and tested on 48 individuals from a single sampling site. Using three multiplex reactions, each primer pair amplified good quality, reliable and polymorphic product. The number of alleles ranged from 9 to 37 per locus and average expected heterozygosities ranged from 0.780 to 0.964. No evidence of linkage disequilibrium was detected between any pair of loci, while significant departure from Hardy–Weinberg expectation was detected at one locus only, suggesting the presence of a null allele at low frequency at this locus. These markers will be employed to measure genetic connectivity and infer influences of connectivity on population maintenance and resilience in this mouth-brooding coral reef fish that inhabits sheltered lagoons in the coral atolls of north–west of Australia.
