Abstract
Notch activation requires unfolding of a juxtamembrane negative regulatory domain (NRR). Tiyanont etal. (2011) analyzed the dynamics of NRR unfolding in the presence of EGTA. As predicted from the crystal structure and deletion analyses, the lin-Notch repeats unfold first, facilitating access by ADAM proteases. Surprisingly, the heterodimerization domain remains stable.
