Abstract

Introduction of computer-assisted semen analysis (CASA) into a laboratory required the development of quality control procedures to ensure proper operation of the method. Results of three experiments designed to compare the performance of a commercial method for CASA to traditional semen analysis techniques are presented. Variances in the measurement of concentration, percent motility, linearity, and velocity were determined, and 95% confidence intervals were established for each parameter (concentration /1.2186, concentration × 1.2186), percent motility ± 10.733%, linearity ± 0.485, and velocity ± 6.87 μ/ sec. The CellSoft system performed reliably with any of three instrument operators. Results of concentration measurements using the CASA system and a traditional hemacytometer method compared favorably in the concentration range of 8.6 to 340 million sperm per milliliter. The CASA method overestimated concentrations below 8.6 million / mL and underestimated concentrations above 340 million / mL.
