Abstract

medium concentrations. This difference was more marked at the low protein concentration. This difference was introduced by the Tecan being programmed to sip very small volumes (smallest 25 pi) of the stock serum albumin, concentrations (2 mg/ml) used to construct the standard curve. Clearly, serial dilutions or sipping larger volumes would eliminate this difference. The interassay reproducibility for the two procedures was equally good (coefficient of variation range 0.29-1.98%). In conclusion, by adjustment of the volumes of reagent added at the two critical stages of the Lowry protein assay further mixing is eliminated. Also, since separate vortex mixing of dilute alkaline solutions is avoided then the automated procedure is safer in use. The Tecan liquid-handling system is quicker than a manual system without any loss in precision. The flexibility of the Tecan liquid-handling system makes the equipment a valuable addition to the automation of routine biochemical assays.
