Abstract

Objective To determine the Sparfloxacin concentration in human plasma. Methods The plasma sample was extracted by dichlormethane and determined by HPLC ,the stationary phase was Gemini C18 (250 mm×4.6 mm, 5 μm), the mobile phase consisted of potassium dihydrogen phosphate-triethylamine-methanol. The flow rate was 1.2 ml·min-1, ultraviolet detector was performed with wavelength detector at 292 nm. Results The retention time of Sparfloxaein was about 9.6 min, and the linear range of quantity was 0.016~2.000 μg·ml-1. The recoveries of method were more than 90%(n=5), RSD<10 % (n=5). Conclusions This method is rapid and accurate, it can be used for the content determination of the plasma Sparfloxacin and the study of pharmacokinetics. Key words: Sparfloxacin; Plasma concentration of drug; HPLC
