Abstract

Objective: To establish a method to determine propofol concentration in human plasma administered with target-controlled infusion.Methods: RP-HPLC method with ultraviolet detector was applied in our study.Separation was performed on a Shim-pack CLC ODS-C18 column with the mobile phase consisting of acetonitrile∶water=65∶35 at a flow rate of 1.5 ml/min.The detection wavelength was 220 nm and the column temperature was 25 ℃.Results: Propofol was shown to be in good linearity within the range of 0.05-5.0 μg/ml(r=0.999 8),with good accuracy and stability.Recovery rates at low,medium and high concentrations(0.5,2.0,5.0 μg/ml) of propofol were(103.0±2.5)%,(98.0±1.4)% and(102.5±3.1)%(n=3),respectively.Conclusion: The method is accurate,sensitive and rapid for the determination of propofol concentration in human plasma administered by target-controlled infusion.
